(a) In pBR322, foreign DNA has to be introduced in tetR region. From the restriction enzymes given below, which one should be used and why :
PvuI, EcoRI, BamHI
 
(b) Give reasons, why the other two enzymes cannot be used.

(a) In pBR322, restriction enzyme BamHI should be used for introducing foreign DNA in tetR region.
 
(b) Pvu I and EcoRI cannot be used for introducing gene in tetR region, because Pvu I is present in ampR region and EcoRI codes for proteins involved in the replication of the plasmid.
